Abstract: The human genome contains ~20,000 genes, but majority of the genome is noncoding. Dissecting cis-regulatory elements hidden in these regions and understanding of the mechanisms by which they exert regulatory effects on specific target genes across distances of kilobases, and in some cases megabases, remain very challenging. I will briefly review our technology development in studying cis-regulatory structures and functional genomics over the years. Then introduce our recent developments in biochemical (BL-Hi-C), biophysical (Super-resolution imaging) and Bioinformatics (Functional DNA loop quantitation and prediction) technologies, in particular on a new methodology CAPTURE -- “In Situ Capture of Chromatin Interactions by Biotinylated dCas9” and its application for in situ capture of individual constituents of the enhancer cluster controlling human β-globin genes, which establishes evidence for composition-based hierarchical 3D regulatory organization and its implication in human diseases.
